Daly enrolled in Columbia University's doctoral chemistry programme, and in 1947 became the first Black woman to earn a PhD in chemistry in the United States, completing her dissertation on the chemical compounds produced during starch digestion under the mentorship of prominent chemist Mary Letitia Caldwell, an achievement of extraordinary significance given the near-total absence of Black women in American chemistry doctoral programmes at the time.
Daly conducted significant research throughout her career on the biochemistry of the human heart and circulatory system, working at institutions including the Rockefeller Institute and Columbia University, and her research contributed directly to scientific understanding of the relationship between cholesterol and clogged arteries, work that provided foundational biochemical evidence supporting the connection between diet, cholesterol and cardiovascular disease that became increasingly significant in subsequent decades of cardiac health research and public health guidance.

Daly established a scholarship in 1988 at Queens College specifically for Black students pursuing physics or chemistry degrees, named in honour of her father whose own chemistry ambitions had been thwarted by financial constraints decades earlier, a deliberate act ensuring other students would not face the same barrier that had originally redirected her father's career and, indirectly, shaped her own determined path into the field.
