Emmett W. Chappelle was born on 24 October 1925 in Phoenix, Arizona, and served in the US Army during World War II before pursuing higher education, earning a degree from the University of California, Berkeley, and a master's degree from the University of Washington, before beginning a scientific research career spanning multiple institutions and disciplines.

Chappelle worked as a research scientist studying photosynthesis and cellular biology, and made a significant scientific discovery establishing that individual bacterial cells could photosynthesise, a finding that expanded scientific understanding of photosynthetic processes at the cellular level and contributed to broader research on food and oxygen production relevant to NASA's interest in supporting long-duration human space travel.

Chappelle joined NASA's Goddard Space Flight Center, where he developed significant expertise in bioluminescence, the biochemical light-emission process found in organisms including fireflies, and applied this research toward developing a method using adenosine triphosphate (ATP) bioluminescence to rapidly detect the presence of bacteria and other microorganisms in a given sample.
